Challenge

The development of the human immune system during early life, including the prenatal period, plays a crucial role in determining susceptibility to various diseases later in life, such as allergies, asthma, and autoimmune disorders. However, the mechanisms underlying immune system imprinting during this critical window are not fully understood, primarily due to challenges in sourcing suitable samples and the complexity of environmental exposures affecting immune development. There is a pressing need to clarify how early-life exposures influence immune maturity and subsequent health outcomes.

Solutions

The INITIALISE project adopts an interdisciplinary approach, integrating data from multiple cohorts and biobanks to investigate the interaction between genetic factors, environmental exposures, the gut microbiome, and immune system development. By employing state-of-the-art technologies in exposomics, metabolomics, and immunology, the project aims to illustrate how genetic and environmental factors influence the gut microbiome, host metabolism, and immune system and how these interactions impact health across the lifespan. The project also seeks to define the role of immune system maturation as a mediator between early-life exposures and long-term health outcomes. Functional studies will be conducted to test specific mechanisms by which environmental factors affect immune development. Additionally, a pilot clinical study targeting the immune system for personalized disease prevention will be implemented.

Results

By deepening our understanding of how early-life exposures shape immune development, INITIALISE aims to inform strategies for disease prevention and health promotion throughout the lifespan. The project’s findings could lead to targeted interventions during critical periods of immune system maturation, potentially reducing the incidence of immune-mediated diseases and improving overall public health.
